Typically, post count builders are people who build forum accounts in hopes of selling the account or signature space to others. These people will usually rush into a forum and make lots of posts within a short period of time. And, once they have created the posts, the signature line is installed. Then the new member is never seen again.
Most real members only create a few new posts a day, unless engaged in some heated on forum debate or conversation. In that situation, it's obvious that the extra posts were made in context. Whereas, a post count builder might make fifty posts in an hour.
Typically though, it's easy to tell when someone is post count building by looking at the quality of the posts.
Does the response relate to the original question? Has the member taken time to actually read the content in previous posts?
A post count builder's response might be very short or generic, sometimes barely relating to the original post.
Another sign of possible post count building are overly simplified newcomer type threads, such as 'What is SEO?' or long articles, which are usually copied from one forum to the next.
